CD44 is expressed in many normal human hematopoietic, epithelial,and mesenchymal cells. Functionally, CD44 is involved in lymphocyte recirculation, adhesion, activation, lymph node homing, and to a larger extent in other tissue types, cell- cell and cell-matrix interactions.1,2,3,4 CD44 is one of two major hyaluronan (HA) binding proteins required for activation of hyaluronan.1,2,3,4 Together, HA and CD44 are involved in enhanced cell signaling and locomotion; both are expressed abundantly during morphogenesis as well as physiological and pathological invasive processes.1,2 The extensive glycosylation patterns of CD44 are important for the recognition and binding of the CD44-hyaluronan complex.5 The CD44 molecule is known to present several different glycosylation, oligomerization, and protein sequence patterns due to differential mRNA splicing.2,3,4 There are 10 variants of CD44 due to differential splicing.3,4 Five of the ten are located in the extracellular domain of the protein.3 Changes in the regulation of the different variant expression patterns in a variety of tissue types has been associated with differing prognosis.3,4,6,7,8,9,10
