L’anticorps CONFIRM anti-Epidermal Growth Factor Receptor (EGFR) (3C6) Primary Antibody de Ventana Medical Systems (Ventana) est un anticorps monoclonal de souris (IgG1) dirigé contre le domaine extracellulaire de la protéine EGFR humaine de 170 kD. L’anticorps reconnaît également la variante de type III de 145 kD. Cet anticorps est conçu pour être utilisé en laboratoire en vue de la détection qualitative par microscopie optique de l’EGFR contenue dans du tissu inclus en paraffine et fixé au formol sur un Ventana Automated Slide Stainer.
L'interprétation clinique de toute coloration ou absence de coloration doit être complétée par des études morphologiques et l'analyse de contrôles adéquats. "Value": "EGFR is a member of the receptor tyrosinekinase Her family. Also known as Her1, EGFR is structurally related to Her2, Her3, and Her4. Members of this family are capable of forming heterodimers to elicitsignal transduction. EGFR is expressed at low levels in most normal epithelial tissues, but can be overexpressed in cancers of the breast, brain, bladder, lung, gastric, head and neck, esophagus, cervix, ovary, endometrium, and particularly in squamous cell carcinomas.1

"Value": "CONFIRM anti-EGFR (3C6) may be used as the primary antibody for immunohistochemical staining of paraffin tissue sections. In general, immunohistochemical staining allows the visualization of antigens via the sequential application of a specific antibody (primaryantibody) to the antigen, a secondary antibody (link antibody) to the primary antibody, anenzyme complex and a chromogenic substrate with interposed washing steps. The enzymatic activation of the chromogen results in a visible reaction product atthe antigen site. The specimen may then be counterstained and cover slipped. Results are interpreted using alight microscope and aid in the differential diagnosis of pathophysiological processes, which may or may not be associated with a particular antigen.
CONFIRM anti-EGFR (3C6) is optimally diluted for use with Ventana detection kits and automated slide stainers. Each step in thestaining protocol includes incubation for a precise time at a specific temperature. Atthe end of each incubation step, the sections are rinsed by the Ventana automated slide stainer to stop the reaction and remove unbound material that would hinder the desired reaction in subsequent steps. To minimize evaporation of the aqueous reagents from the specimen containing slide a coverslip solution is applied in the slide stainer.
