High Resolution Melting

Minimum Sample Number
A minimum of 5 samples is required for an analysis by the automated grouping algorithm.
If 3 or more groups are expected the mandatory minimum requirement is 7 samples.

Maximum number of acquisition
The instrument supports a maximum number of 2000 measurements for one HRM run.
This number can't be exceeded in a \"normal\" HRM run, even when the highest acquisition rate of 25/°C is used.
However, there might be applications out of range that include measurement in multiple channels that could exceed the maximum of 2000 measurements.
In this case we advise to either reduce the temperature range or the acquisistion rate. The calculation for the total number of measurements (formula) is:

Importing MP24 sample lists
Although not mentioned in the current LightCycler® 96 Operators Guide, the Application Software also allows for the import of MagNA Pure 24 sample lists (.txt or .xml) in addition to MagNA Pure 96 sample lists.
The import can be achived via the option Sample Editor -> Table View -> Import  MP96 Sample List.
So essentially MP24 sample list can be imported via the option that is also used to import MP96 sample lists.
Frequency of restarting the LightCycler 96 instrument
It is strongly recommended to restart the LightCycler 96 instrument once a day to avoid potential software problems even if the instrument is not used.

SMBv1 network protocol required for Automated Backup to Network Share feature
The Automated Backup to Network Share feature from LightCycler® 96 uses a Network Protocol called Server Message Block (SMB) also called \"samba\".
Several version of the SMB protocol exists and are used.
The LightCycler® 96 Instrument is only compatible with the version 1 of the protocol (SMBv1).
Disabling or blocking SMBv1 on the Network will affect the Automated Backup feature.
So customers using the Automatic Backup feature must allow the SMBv1 protocol on their Network.

Instrument Software V1.2

Units Concerned

Instrument Software V1.2 will be pre-installed on all instruments equippted with the new IPC (Internal PC)-Baytrail.
All LightCycler® 96 with Serial Number 13426 and higher that are produced with the new IPC (Internal PC)-Baytrail including the new Instrument Software V1.2.
Older Instruments will nnot and can't be updated to the new software.

Executive Summary
The software version 1.2:
  • Does not include new features or bug fix
  • Does not change the specification of the instrument
  • Is not backwards compatible
  • Can only be used in combination of IPC-Baytrail
  • Is available on GRIPS (see below for details concerning the end-to-end protection)
  • Includes minor changes in the GUI
  • Has additional logs for the Operating System
  • Contains a few limitation

GUI changes
1. Introduction of an \"About\" screen.
2. The USB logo is blinking red when a USB device is in use and should not be disconnected.
3. Design of the startup splash screen has changed and is now composed of the following screens.
4. The start sequence shows the additional step \"Startup\".

Other non-visible changes
1. Roche Vanilla Agent for Axeda is updated to version
2. The underlying Linux OS was updated.
3. The zip file created for the logs has a new folders structure
  • ic folder contains Instrument Control logs: No change in the content
  • im folder contains Instrument Management logs and error logs: No change in the content
  • os folder contains Linux OS and SW update logs: New logs for 2nd and 3rd level support

The Instrument Software version 1.2 is available on GRIPS in the folder \"Instrument Software Download\" and is also located on the USB drive delivered with the instrument. The package can only be used to reinstall the software on the instrument.
Installing the Software version 1.2 on a LightCycler® 96 Instrument with IPC Small is not possible.
The software is protected by an integrated end-to-end protection.
The instrument will not install the package if it was altered.

The current \"LightCycler 96 System Manual 2.0 SW 1.1 and Addendum 1 - June 2016\" also called System Guide, is valid for Instrument Software version 1.2. The System Guide contains the following statement: \"Roche Diagnostics GmbH reserves the right to make any changes necessary without notice as part of ongoing product development.\" Moreover there were no changes in the specification

The instrument software V1.2 has the following known limitation:
  • It is not possible to access the instrument on the network by using the hostname. The IP address of the instrument must be used.
  • It is not possible to recalibrate the screen (re calibration of the screen is lost after reboot)

A corresponding \"Declaration of Software Validation\" has been released and has been uploaded under \"Downloads\"-\"Certificate\".

Workaround for Statistical Issue in Relative Quantification Dual Color Assays

For a Dual Color assay setup with 2 Reference results for every sample, an error calculation is done even if no replicates are present in the run if the same sample name is used for 2 different target genes.
For this particular case, the customer can avoid this by naming the samples differently. Later, the customer can then create 2 Analysis, one for each gene This is the only way the software can be prevented from making a replicate statistic in case of a Dual Color application in case of more than one target gene per sample but identical reference genes for both targets.

Display issue for for certain rarely used dye combinations affecting the Endpoint Genotyping Application of Software Version 1.1

The results of the Endpoint fluorescence analysis are not correctly displayed, whenever the second dye name used in the analysis is earlier in alphabetical order than the first dye name.
For all of this dye combinations (dye A - dye B), the results are displayed switched between the dyes, so that:

  • all wells called \"Homozygote for dye A\" are in reality \"Homozygote for dye B\" and vice versa
  • the scatterplot axis labeled as \"dye A\" is the axis for \"dye B\" instead and vice versa
  • the homozygote heatmap entries are switched between the dyes

The following very rarely used dye combinations are impacted by the issue:

  • FAM - Cy5
  • Hex - Cy5
  • Red610 - Cy5
  • TexasRed - Cy5
  • VIC - Cy5
  • VIC - Red610
  • Yellow555 - Texas Red
  • Yellow555 - Red610
  • Yellow555 - Cy5

The most commonly used dye combination (FAM - VIC) and all other dye combinations listed below are NOT impacted by the issue:

  • FAM - Hex
  • FAM - Red610
  • FAM - TexasRed
  • FAM - VIC
  • Hex - TexasRed
