"Value": "Red Counterstain II contains a cellular dye, nuclear fast red, which forms chelates with aluminum cations, generating a pink color associated with cellular components.1 During the in situ hybridization (ISH) process, hapten labeled probes are bound to specific target sequences in cells or tissue. This binding is visualized by an alkaline phosphatase based detection system, which generates a visible blue precipitate. As a contrast, Red Counterstain II provides pink background staining in nuclei and cytoplasm of tissue sections to assist with the slide reading under bright field microscope.

"Value": "In general, in situ hybridization (ISH) staining allows the visualization of target DNA or RNA sequences via the sequential hybridization of a labeled DNA or RNA probe to the target, a primary antibody against the labeled probe, and a secondary antibody to the primary antibody, an enzyme complex and a chromogenic substrate with interposed washing steps. The enzymatic activation of the chromogen results in a visible reaction product at the target site. At the end of the incubation step, the instrument washes the slides to stop the reaction and remove unbound material.
For ISH reactions that result in a visible blue precipitate, the slides may be counterstained with Red Counterstain II.
