DNA library preparation for next-generation sequencing (NGS) involves the fragmentation of target DNA molecules into varying sizes. The KAPA library prep portfolio offers kits for both enzymatic fragmentation and mechanically fragmented DNA. The fragmented DNA is then subjected to end repair/A-tailing and ligation of platform-specific adapters to the library.

The libraries are subsequently amplified (in with-PCR workflows) and cleaned up using beads (this may also include size-selection steps). Depending on the nature of the DNA sample–for example, a fresh sample versus a formalin-fixed paraffin-embedded (FFPE) tissue–the workflow may require optimization.
