High-quality sample preparation accessories for high-performance NGS

All NGS library preparation workflows—whether for DNA or RNA—require numerous steps, including the addition of sequencer-specific adapter sequences and at least one bead-based purification step. Target-enriched NGS workflows require additional NGS accessories such as oligos (probes and primers), amplification reagents, capture beads, and washing reagents.

The quality of these NGS accessories is paramount, even when starting with the highest-quality reagents in the core library construction kit. For example, incorrect adapter indices or low-efficiency bead-based steps may result in the loss of precious samples—or even the failure of entire sequencing runs.

We supply all of the sample preparation accessories needed for each of our DNA and RNA library construction workflows, including target enrichment probes, primers, and accessories. The full portfolio includes:

  • Adapters for DNA and RNA library preparation:  full-length KAPA UDI Adapters (96 adapters with UDI unique dual indexes included); KAPA Universal Adapter or KAPA Universal UMI Adapter (to be used in combination with KAPA UDI Primer Mixes 1-384, which add the index sequences); KAPA Adapter Dilution Buffer.
  • Beads for library prep and cleanup: KAPA HyperPure Beads and KAPA PureBeads.
  • Capture reagents for hybridization-based target enrichment: KAPA HyperCap probes,  KAPA HyperCapture Reagent Kit, KAPA HyperCapture Bead Kit, KAPA Probes Resuspension Buffer, KAPA Universal Enhancing Oligos (UEOs), and KAPA Hybrid Enhancer Reagent.

While all suppliers subject their reagents to quality control (QC) testing, not all suppliers also validated each reagent across their workflows or use NGS-based QC. All of our NGS accessory reagents are validated in Roche sample preparation and target enrichment workflows to ensure index accuracy, optimized yields, and high success rates across all sample types and applications.

  • Adapters undergo sequencing-based QC testing to confirm their barcode sequences and minimize the risk of index misassignment arising from barcode cross-contamination
  • All batches of adapters and beads are carefully validated to ensure optimal yields and performance

The type and amount of adapters and beads required for a library preparation workflow may differ widely, depending on the input material, fragment length (size selection requirements), target enrichment strategy (if applicable), level of multiplexing, and sequencing application. To best support our entire portfolio of DNA and RNA library construction and target enrichment workflows, NGS accessories for sample preparation are available as standalone kits in a variety of formats.

  • Adapter options include universal, truncated adapters with or without unique molecular identifiers (UMIs) or full-length unique-dual indexed adapters (UDI)
  • Pack sizes come in a variety of sizes to support different inputs, multiplexing strategies, and applications while ensuring minimal wastage
  • Reagent formats include automation-friendly plated reagents or tubes, as well as bulk bead formats.

*The sets of 192 and 768 barcode sequences used in KAPA UDI Adapters and KAPA UDI Primer Mixes (respectively) are exclusive to Roche and different between the two sets. All barcodes are compatible with all Illumina instruments with patterned or non-patterned flow cells. Note that KAPA Adapters are not compatible with methyl-seq applications.

**This is sufficient for four or one library preps (KAPA UDI Adapter plate or KAPA UDI Primer Mixes plate, respectively) with the KAPA EvoPrep or KAPA EvoPlus V2 kits if no adapter dilution is required. Generous overfill supports use on automated liquid handling systems.

†KAPA UDI Primer Mixes and KAPA Adapter plates are shipped with peelable seals. Replacement seals (three per plate) are peelable and pierceable.
