In many biomedical research settings, targeted next-generation sequencing (NGS) offers several advantages over whole-genome sequencing (WGS). For example, the use of NGS panels for enrichment of specific genomic regions can:

  • Reduce sequencing costs while enabling deeper sequencing, as specific regions of interest can be interrogated with greater depth for higher sensitivity
  • Simplify both computational analysis and data storage, as NGS panels yield more focused data and less data from other genomic regions.

However, NGS panel design can be complex and time-consuming, requiring several rounds of design and experimental trial-and-error to sufficiently and consistently cover the desired region. To make it easier for researchers to get started with targeted NGS without designing new panels, Roche offers predesigned NGS panels for both of our NGS target enrichment workflows: the hybridization-based KAPA HyperCap Workflow and the KAPA HyperPETE workflows, which uses primer-extension target enrichment.

We offer two types of predesigned panels to reduce the effort and expense needed to perform targeted NGS:

  • Catalog Panels - designed by expert scientists, with in-house analytical validation, and supplied from stock
  • Design Share Panels - developed in collaboration with leading scientists around the world, and customizable through the use of Roche’s HyperDesign Tool

And, if you do want to design your own panels, the online HyperDesign Tool is a free, easy-to-use, flexible NGS panel design tool that lets you create your own panels from scratch.

While panel design is critical to ensure that all of the genes/regions of interest are uniformly covered, probe quality is just as important in ensuring robust and reproducible sequencing results. 

Predesigned Roche sequencing panels are extensively tested, either in-house at Roche and/or by external collaborators. This ensures high on-target rates and coverage metrics, as well as accurate and sensitive mutation detection. 

Every batch of Catalog Panel probes is subjected to NGS-based quality control (QC) testing to confirm probe presence and concentration.

Predesigned, validated NGS panels reduce implementation and ordering times

The time required to design, optimize, validate, and implement a new NGS panel can range from weeks to months, and probe manufacturing can add days to weeks to the total turnaround time if an existing panel is not available in the lab. 

By selecting a predesigned Catalog or Design Share Panel, the time from panel design to implementation can be reduced significantly. Catalog Panels are supplied from stock. Therefore, lead times are similar to those for the other reagents required for your KAPA HyperCap or KAPA HyperPETE workflow. 

Single-vendor support for the entire workflow simplifies the ordering process and inventory management.
