The test utilizes amplification of target DNA by RT-PCR and nucleic acid hybridization for the detection of CT and differentiate the strains (L1, L2a, L2b and L3) causing Lymphogranuloma venereum (LGV) from non LGV strains. en Bacterial DNA is analyzed by real-time PCR, that amplifes a 79 bp long fragment of polymorphic membrane protein H (pmpH) gene, in channel 02 using a FAM-labelled probe for CT non LGV and channel 03 using a HEX-labelled probe specific for CT invasive serovars L1, L2a, L2b and L3 (LGV) strains. The (+) Ctrl comprises plasmids with a 331 bp long synthetic fragment of pmpH gene from a LGV strain, and a 401 bp long synthetic fragment of pmpH gene from a non-LGV strain (Accession number: MG542649 position 3 - 334 or 3 - 405 respectively). en The limit of detection (LoD) for UC-TIB-LGV was determined by analysis of serial dilutions of plasmid DNA in cobas® PCR media (400 μL sample volume).
