The test utilizes amplification of target RNA by RT-PCR and nucleic acid hybridization for the detection and discrimination of the arthropod-borne Zika virus (ZikV), Chikungunya virus (ChikV), Dengue virus (DenV) serotypes 1-4 and West Nile virus (WNV). In Addition to WNV lineage 1 and 2, Japanese encephalitis virus, St. Louis encephalitis virus, Murray Valley encephalitis virus, Usutu virus and Kunjin virus (WNV variant) are detected as well. All these Flaviviruses belong to the Japanese encephalitis sero-complex and show nucleotide sequence homology with WNV. en The limit of detection (LoD) for UC-TIB-Tropical-Virus was determined by analysis of serial dilutions of the WHO International Standards for ZikaV (11468/16), ChikV (11785/16), DenV-2 and WNV (18/206) obtained from PEI, FDA and NIBSC respectively. Diluted in virus-negative human plasma (850 μL sample volume). The study demonstrates that the UC-TIB-Tropical-Virus detects ZikaV RNA at a concentration of 60 IU/mL, ChikV and Denv-2 RNA at a concentration of 10 IU/mL and WNV RNA at a concentration of 20 IU/mL respectively, determined by LOGIT analysis with a hit rate of 95%. en Viral RNA is analyzed by real-time PCR that amplifies a 134 bp long fragment of E gene from ZikV detected with a Coumarin-labeled probe in channel 01, a 94 bp long fragment of E1 gene from ChikV detected with a FAM-labeled probe in channel 02, a 130 bp long fragment of the 5 ́-UTR from DenV detected with HEX- labeled probes in channel 03 and a 236 bp long fragment of the 3 ́-UTR from WNV analyzed with a LC640 labeled probe in channel 04. The (+) Ctrl comprises plasmids with a 275 bp long synthetic fragment of E gene from ZikV, a 3056 bp long synthetic fragment of E1 gene from ChikV, a 277 bp long synthetic fragment of 5 ́-UTR from DenV-2 and a 476 bp long synthetic fragment of 3 ́-UTR from WNV (Accession number: NC_012532 position 1076 - 1347 for E gene from ZikaV, DQ443544 position 8704 - 11696 for E1 gene from ChikV, MK506263 position 2 - 280 for 5 ́-UTR from DenV-2 and HM051416 position 10530 - 10981 for 3 ́-UTR from WNV-1 respectively). en
