This workflow contains all of the buffers and enzymes required for the depletion of ribosomal RNA (rRNA) and the rapid construction of stranded RNA-seq libraries from purified total human RNA. This workflow depletes both cytoplasmic (5S, 5.8S, 18S, and 28S) and mitochondrial (12S and 16S) rRNA species. Accurate strand origin information is maintained using dUTP incorporation during 2nd strand synthesis. This workflow is applicable to a wide range of RNA-Seq applications, including: Gene expression analysis of high- and low-quality RNA samples Single nucleotide variation (SNV) discovery Splice junction and gene fusion identification Characterization of both polyadenylated and non polyadenylated RNAs, including noncoding and immature RNAs. Final DNA libraries generated from this workflow should be pooled manually. The final library pools should be run on an Illumina sequencing platform, such as the Illumina® NovaSeq™ 6000, or other suitable systems. This workflow uses the AVENIO Edge KAPA User-Filled Kits (KUFK). Reagents from the manual reagent kit are pipetted into the respective KUFK plastics that are specific to the RNA workflow. en
