Congo Red Buffer is added to differentiate the stain. Special Stains Hematoxylin, a Mayer’s hematoxylin solution, is used to provide contrasting blue to purple nuclear staining. The β-pleated sheets of amyloid are suitable in size and shape to accommodate the Congo red molecules, which are held in the latticework of the β-pleated sheets.6 Birefringence under polarized light is an intrinsic property of the amyloid fibril Congo red complex.7,8This kit is optimized for use on BenchMark Special Stains instruments.
