Periodic Acid 1% is used to oxidize carbohydrates to aldehyde groups. The combined Silver Nitrate 1% and Silver Buffer-(Triethanolamine <5%) solutions form a triethanolamine-silver complex that is easily reduced to metallic silver by the aldehyde groups. To enhance staining contrast, toning of the tissue specimen is completed using gold chloride within the Gold Chloride <0.5% reagent. Sodium Thiosulfate 2% stops the reaction and removes any unreduced silver and gold from the section.5,8This kit is optimized for use on BenchMark Special Stains instruments. The reagents are applied to tissue on microscope slides and mixed over the entire specimen.5. Bancroft JD, Layton C. Connective and Other Mesenchymal Tissues with Their Stains.
