The mutation site in each PCR fragment is analyzed using labeled probes. Tube 1 amplifies and detects G719X, exon 19 deletions, C797S, T790M and L858R mutations as well as a wild-type reference sequence on exon 19. Tube 2 amplifies and detects exon 20 insertions, L861Q, S768I as well as a wild- type reference sequence on codon 768. An 81 bp fragment covering the EGFR G719X mutations is amplified with specific primers. The mutation site in the PCR fragment is analyzed using three mutant probes labeled with Atto-425. A 113 bp fragment covering the EGFR C797S and T790M mutations is amplified with specific primers. The mutation site in the PCR fragment is analyzed using three probes. The probes for exon 20 insertions are FAM-labeled, and the probe for S768I is Texas Red-labeled. A wild-type reference sequence is also detected on codon 768 using a Cy5.5 labeled probe. A 76 bp fragment covering the EGFR L861Q mutations is amplified with specific primers. The mutation site in the PCR fragment is analyzed using one mutant probe labeled with HEX. The two PCs included in the kit contain 2-10 % synthetic mutant targets in wild-type genomic DNA and are equivalent to ~17 ng/µL when dissolved in 60 µL of PCR grade water. It is recommended to always include at least one PC and one No-Template Control (NTC; e.g. PCR grade water) in each run per PSR.
