LoD not tested. en Real-time PCR is employed to analyze human DNA for HFE gene mutations. To detect the p.C282Y (c.845G>A) polymorphism, a 103 bp fragment is amplified. The wild-type p.C282 (c.845G) is detected in channel 01 using a Coumarin-labeled probe, and the p.C282Y mutation (c.845G>A) is detected in channel 04 using an LC640-labeled probe. Concurrently, to detect the p.H63D (c.187C>G) polymorphism, a 93 bp fragment of the HFE gene is amplified.
