This product detects TBEV including other members of the TBEV serocomplex like the Oms Hemorrhagic Fever Virus (OHFV), Louping Ill Virus (LIV), Langat Virus (LaV) and Powasan Virus (PowV). Kyasanur Forest Disease Virus (KFDV) and Alkhurma Virus (AlkV) are not amplified, based on in-silico analysis. The Borrelia spp. detected, include Borrelia burgdorferi sensu stricto, B. miamotoi, B. afzelii, B. spielmanii, B. garinii, B. lusitaniae and others. en The limit of detection (LoD) for UC-TIB-Tickborne-1 was determined by analysis of serial dilutions of plasmid DNA in cobas® PCR media (400 μL sample volume). The study demonstrates that the UC-TIB-Tickborne-1 detects at least 32 genome equivalent copies or less per mL for TBEV, 18 copies or less per mL for Borrelia, 35 copies or less per mL for Bartonella and 30 copies or less per mL for C. burnetii, determined by LOGIT analysis with a hit rate of 95%. en For TBEV RNA a 67 bp amplicon from the 3 ́-UTR is amplified and detected with Coumarin-labeled probes in channel 01. Borrelia spp. DNA is analyzed amplifying a 74 bp long fragment of the 23S rRNA gene with a FAM-labeled probe in channel 02, for Bartonella spp. a 74 bp long fragment of the gltA gene is amplified and detected with a HEX-labeled probe in channel 03 and for C.burnetii a 103 bp long fragment of the IS1111a gene is amplified and detected with a LC640 labeled probe in channel 04. The (+) Ctrl comprises plasmids with a 68 bp long synthetic fragment of 3 ́-UTR of TBEV, a 403 bp long synthetic fragment of 23S rRNA gene of Borrelia spp., a 142 bp long synthetic fragment of gltA gene of Bartonella spp. and a 1041 bp long synthetic fragment of IS1111a gene of C. burnetii (Accession number: AH002418 position 756 - 823 for 3 ́-UTR of TBEV, NR_076348 position 585 - 990 for 23S rRNA gene of Borrelia spp, MN107415 position 280 - 421 and M80806 position 78 - 1110 for IS1111a gene of C. burnetii respectively). en
