KAPA EpiVault EvoPrep Kit
Protect the signal. Shorten the path.
The KAPA EpiVault EvoPrep Kit is a high-performance library preparation solution engineered to minimize methylation bias (mBias) and maximize data accuracy from low-input cell-free DNA (cfDNA). By pairing the new methylation-bias reducing KAPA EpiVault module directly with the proven KAPA EvoPrep Boost chemistry, this combined workflow preserves critical biological signals from input amounts as low as 1 ng.1 With a streamlined protocol completed in just ~70 minutes (without PCR) using only four total tubes and minimal extra pipetting steps, it seamlessly delivers richer, high-confidence methylation insights without adding workflow complexity.1
mBias is an important metric that we talk about here, but what does it really mean? Methylation bias (mBias) refers to systematic, artificial distortions in reported DNA methylation levels at specific positions along sequencing reads—most prominently at the extreme 5′ and 3′ ends. Rather than reflecting true biological methylation states, these artifacts arise during library preparation. Standard double-stranded DNA (dsDNA) library construction relies on end-repair and A-tailing (ER/AT) steps, where enzymatic fill-in using unmethylated cytosines (or end degradation) artificially skews the terminal bases toward zero or reduced methylation signal.2,3
Left uncorrected, mBias introduces technical noise that distorts bioinformatic downstream processing. In research applications requiring precise, quantitative methylation profiling—such as liquid biopsy cell-free DNA (cfDNA) tissue-of-origin deconvolution and epigenetic biomarker discovery—even minor localized artifacts can cause misclassification or mask subtle biological signals.3,4 By eliminating ER/AT-induced mBias at the chemistry level, researchers can preserve maximum read utility without resorting to bioinformatic trimming that discards valuable sequencing coverage.
